Benomile bağlı subkkronik karaciğer hasarı ve fibrogenezine karşı kondroitin-4-sülfat (C4S) ve ?-lipoik asitin (LA) koruyucu etkilerinin araştırılması

Özet
Bu çalışmada, fungusit olarak kullanılan benomile bağlı oluşan subkronik karaciğer hasarı ve fibrosise karşı kondroitin-4-sülfat (C4S) ve ?-lipoik asit (LA)' in koruyucu etkileri incelendi. Benomil (200 mg/kg), ?-lipoik asit (200 mg/kg) ve kondroitin-4-sülfat (25 mg/kg) 5 hafta boyunca haftada bir kez olmak üzere intraperitonal (IP) injeksiyon yoluyla uygulandı. LA ve C4S benomil uygulamasından 30 dakika önce uygulandı. Deney süresi sonunda ölçülen vücut ağırlıklarında, benomil grubundaki hayvanlarda önemli bir değişiklik olmaz iken, antioksidan uygulanan hayvanlarda istatistiksel olarak anlamlı bir artış (p<0.01, p<0.02) tespit edildi. Hepatik hücre hasarının belirlenmesi için karaciğerde miyeloperoksidaz (MPO) enziminin aktivitesi ölçüldü; Benomil grubundaki hayvanların karaciğer dokusu MPO aktivitesinin önemli olarak arttığı (p<0.001), buna karşılık C4S (p<0.05) ve LA uygulanan hayvanlarda MPO aktivitesinin kontrol düzeylerine geri döndüğü (p<0.01) gözlendi. C4S ve LA' nın birlikte uygulandığı grupta azalma daha fazladır (p<0.05). Benomil uygulanmış hayvanların karaciğerlerinde fibrosis oluştuğu histolojik olarak ekstraselüler matriks ve sinüzoidal alanlarda bağ doku artışı ile gösterildi. C4S ve LA uygulamasının kollagen fibril oluşumunda artışa neden olmadığını göstermiştir. Sonuç olarak, antioksidan özellikleri bilinen C4S ve LA' nın, benomile bağlı olarak oluşan karaciğer hasarına ve fibrozise karşı da etkili oldukları söylenebilir.
Abstarct
In this study, the protective effects of ?-lipoic acid and chondroitin-4-sulphate, against subcronic liver damage and fibrosis as a result of benomyl, which is used as fungucide, were examined. Benomyl (200 mg/kg), ?-lipoic acid (200 mg/kg) and chondroitin-4-sulphate (25 mg/kg) are applied by intraperiotenal injection once in each week for a period of 5 weeks. it was determined that body weigths of animals in the groups of control, benomyl+ ?-lipoic acid, benomyl+ chondroitin-4-sulphate and benomyl+?-lipoic acid+ chondroitin-4-sulphate were significantly increased although the body weights of animals in the group of benomyl didn't change considerably. Myeloperoxidase activities were found to be significantly increased in benomyl group when compared with control group . These enzyme activities were observed to significantly decrease in benomyl+?-lipoic acid, benomyl+ chondroitin-4-sulphate and benomyl+?-lipoic acid+ chondroitin-4-sulphate groups when compared with benomyl group. It was determined that when C4S and LA were injected in the rats, they didn't cause an increase in kollagen fibril appearance. It was observed that fibrosis appeared in the liver of rats injected benomil and connective tissue increased in extracellular matrix and sinusoidal areas. The protective effects of C4S and LA against subcronic liver damage and fibrosis as a result of benomyl which is used as fungucide, were examined. Consequently, ıt can be said that C4S and LA, which are antioxidan, are effective against fibrosis and liver damage as a result of benomyl
