Özet
Bu çalışmada Edirne Bölgesi civarında yetişen Trifolium resupinatum L. var. microcephalum Zoh. bitkisinin fitokimyasal incelemesi yapılmıştır. Bitki toplandıktan sonra gölgede kurutulmuştur. Kurutulan bitki CH2Cl2 ve EtOH ile maserasyon tekniğine göre ekstraksiyon yapılmış ve elde edilen ekstraktlardaki çözücüler evaporatörde uçurulmuştur. EtOH ekstresi su ile sulandırılarak EtOAc ile ekstrakte edildi. CH2Cl2 ve EtOAc ekstresine kolon kromatografisi, ince tabaka kromatografisi, preparatif ince tabaka kromatografisi uygulanarak, sekonder metabolitleri ayırma işlemleri gerçekleştirilmiştir. Tek maddelerin saflaştırılma işlemlerinde preparatif ince tabaka kromatografisi ve Sephadex LH-20 kullanılmıştır. Her iki ekstreden toplam 10 bileşik izole edilmiştir. CH2Cl2 ekstresinden 3 tane hidrokarbon, 1 tane steroid, 1 tane triterpen, 1 tane kumarin, 1 tane izoflavon olmak üzere toplam 7 bileşik izole edilmiştir. Etilasetat ile tüketilen kısmından 2 tanesi izoflavon glukozit, 1 tane flavonol glikozit olmak üzere toplam 3 bileşik izole edilmiştir. Bitkiden izole edilen maddelerin yapıları, UV, IR, 1D ve 2D NMR teknikleri ve Mass spektrometresi kullanılarak kimyasal yapıları açıklanmıştır. CH2Cl2 ekstresinden elde edilen 3-metil-1-nonen-3-ol bileşiği ilk defa bu çalışma sonucu bir bitkiden izole edilmiştir.
In this study, Trifolium resupinatum L. var. microcephalum.Zoh. which grows in Edirne zone have been phytochemically investigated. First the plant was collected and dried. Then dry plant was extracted by maceration with CH2Cl2 and EtOH respectively and extracts were concentrated in an evaparator it vacum. Ethanolic extracts was extracted with EtOAc after diluted with water. The separation process of secondary metabolites were carried out from both extracts by using the coloumn chromatography, thin layer chromatography and preparatif thin layer chromatography methods. Preparative thin layer chromatography and Sephadex LH-20 methods were used to purify compounds. Totally 10 compounds were isolated from CH2Cl2 and EtOH exctract. The 3 hydrocarbons, 1 steroid, 1 triterpene, 1 coumarin, 1 isoflavon compound were isolated from CH2Cl2 extract. 2 isoflavon glucosides and 1 flavonol glycoside were isolated from EtOAc extract. The chemical structure of isolated compounds from the plant have been elucidated by using UV, IR, 1D and 2D NMR techniques and mass spectroscopic data. In this study, 3-metil-1-nonen-3-ol compound was obtained from Trifolium resupinatum L. var. microcephalum for the first time.
