Nonıyonik surfaktant “ triton X-114” ve katyonik surfaktant “gemini 10-2-10”un soğan (Allium Cepa L.) ve mercimek (Lens Culinaris Medik ) üzerindeki genotoksik etkilerinin belirlenmesi

Özet
Bu çalışmada, yaygın olarak kullanılan oktifenol serilerinden noniyonik surfaktant "Triton X-114" ve katyonik surfaktant "Gemini 10-2-10", genotoksik etkilerini incelemek için seçildi. Surfaktantlar, soğan (Allium cepa L.) ve mercimek ( Lens culinaris Medik) bitkileri kullanarak laboratuvar koşullarında değerlendirildi. Kökler üzerindeki fitotoksik etkiler 7 gün sonunda belirlendi. Fitotoksisite, kök uzunluğu ve tohum çimlenme yüzdesi üzerindeki fitotoksik etki ,% 50 (EC50) değeri saptandı. Genotoksisite sonuçları test materyallerindeki, mitoz bölünme evreleri, mitotik indeks ve kromozom aberasyonlarına bakılarak belirlendi. Test materyalleri ışık mikroskobu altında incelenerek ve fotoğraflar Olympus fotomikroskobu tarafından alındı. Hücre döngü analizi için ise Flow sitometri yöntemi kullanıldı. Deneyler en az 3 kez tekrarlanarak ve sonuçlar istatiksel olarak değerlendirildi.
In this study, two widely used octylphenol series non-ionic surfactant Triton X-114 and cationic surfactant "Gemini 10-2-10"the gemini surfactants chosen to examine their genotoxic effect. They was evaluated under laboratory conditions using onion (Alium cepa L.) and lentil (Lens culinaris Medik) plants. The phytotoxic effects on root determined after 7th days. Surfactants used in study tested at different surfactant concentrations. The phytotoxicity results was based on the effective concentration that reduced root growth and seed germination by 50 % (EC50). The genotoxicity results was based, the phases of mitosis, mitotic index and chromosomal abnormalities in test materials. The test materials was examined under a light microscope and photogral was taken by an Olympus photomicroscope. The cell cycle analysis performed by flow cytometry method. The experiments were repeated 3 times and the statistical analysis was performed acoording to these results.
