MISIR VE MISIR ÜRÜNLERİNDE TOPLAM FUMONISIN VARLIĞI VE BEBEK FEKESİNDEN İZOLE EDİLEN LAKTİK ASİT BAKTERİLERİ ÜZERİNE FÜMONİSİN Bı'İN ETKİSİ Doktora Tezi Sibel ÖZÇAKMAK TRAKYA ÜNİVERSİTESİ FEN BİLİMLERİ ENSTİTÜSÜ GIDA MÜHENDİSLİĞİ ANADİLİM DALI ÖZET Bu araştırmada farklı bölgelerden toplanan jmşjr. ve mısır, jörjürjerinde toplam fiımonisin varhgL ve bebek fekesinden izole edilen laktik asit bakterileri üzerine fumonisin Bı (FBı)'in etkisi araştırılmıştır. Toplam 56 adet dane mısır örneklerinin (19'u yaş, 37'si kuru) 41 'inde (%73,21'i) toplam fumonisin miktarı 1,7-429,8 mg/kg aralığında ve ortalama 40,81 mg/kg olarak tespit edilmiştir. Toplam 40 adet koçan mısır örneğinin 29'unda (%72,5) 0,2-234,6 mg/kg arasında ve ortalama 40,04 mg/kg toplam fumonisin belirlenmiştir. Analiz edilen 33 adet öğütülmüş mısır ürünlerinin (un, nişasta, irmik ve kırma) %90,91'inde 0,8-425,5 mg/kg arasında ve ortalama 61,9 mg/kg toplam fumonisin belirlenmiştir. Toplam 13 adet mısır çerezi ve mısır cipsi örneklerinin %38,46'smda (5 adet) fumonisin bulunamamış, %61'inde 0,5-37,2 mg/kg arasında ve ortalama 16,23 mg/kg fumonisin belirlenmiştir. Fumonisin Bı'in (FBı) laktik asit bakterileri (LAB) üzerine etkisini tespit etmek için, fumonisin bulunmayan, 1,25 mg/L ve 2,5 mg/L FBı içeren besin ortamında 6 bakteri izolatının 48 saatlik inkübasyon süresince gelişmeleri incelenmiştir. Araştırmada L. rhamnosus'a. ait 3 izolat (IF-I, IV ve V), L. paracasei subsp. paracasefye ait 2 izolat (IF-II ve VI) ve L.fermentum'a. ait 1 izolat (İD) kullanılmıştır. FBı 'e en duyarlı bakteri suşlan IF-I ve II olurken, en dirençli suşlann IF- VI ve IV olduğu belirlenmiştir. THE PRESENCE OF FUMONISINS IN CORN AND CORN BASED PRODUCTS AND THE EFFECTS OF FUMONISIN B! ON LACTIC ACID BACTERIA ISOLATED FROM BABY FAECES Doctorate Thesis Sibel ÖZÇAKMAK TRAKYA UNIVERSITY GRADUATE SCHOOL OF NATURAL AND APPLIED SCHCNCES DEPARTMENT OF FOOD ENGINEERING SUMMARY In this study, the presence of total fumonisins in com and corn products taken from different regions and the effect of fumonisin Bı (FBi) on lactic acid bacteria isolated from baby faeces were investigated. In the 41 (73.21%) milled corn samples out of 56 (19 of them: moist, 37 of them: dried), the quantity of total fumonisins was determined between 1.7-429.8 mg/kg and the average value was 40.81 ppm. In 29 (72.50%) corncob samples out of 40, 0.2 - 234.6 mg/kg total fumonisin with an average value of 40.04 mg/kg was determined. In the 90.91% of the 33 milled corn products (flour, starch, semolina and grained) samples analyzed, 0.8 - 425.5 mg/kg fumonisin with an average value of 61.9 mg/kg was determined. In the 38.46% (5 samples) of the 13 cornflakes and cips samples, fumonisin wasn't determined, and in 61.54% of them, 0.4 - 37.2 mg/kg fumonisin with an average value of 16.23 mg/kg was determined. In order to determine fumonisin Bi (FBi)'s effect on growth of six lactic acid bacteria isolates (LAB) isolates in different media (without FBI, with 1.25 mg/L FBi and with 2.5 mg/L FBi) during 48 hours incubation period was examined. In this research three isolates of Lactobacillus rhamnosus (IF-L V and two isolates of L. paracasei subsp. paracasei (IF-H, VI) and one isolate of L. fermentimi (IF-DI) were used. It was determined that the isolates IF-I and II were most susceptible against FBi and the isolates IF-IV and VI were the most resistant against FBi.
