Ağır metal miks uygulanan vasküler düz kas hücrelerinde antioksidan sistem ve apoptoz yolağı arasındaki moleküler etkileşimler

Özet
Ağır metaller en yaygın kirletici maddeler olup, özgül ağırlıkları 5 g cm-3 den fazla olan maddelerdir. İnsanların ağır metallere maruziyeti en fazla ağır metal içeren yiyecek ve suların tüketimi ile olmaktadır. İçme suyu ile alınan metaller vücutta genotoksisite ve çeşitli hastalıklara neden olabilmektedir. Bu nedenle insani tüketim amaçlı kullanılan sularda izin verilen ağır metal limitleri yasa ve yönetmeliklerle belirlenmiştir. Çalışmamızda insan sağlıklı aorta düz kas hücrelerinde (TG-HA-VSMC) içme sularında izin verilen limitlerde metal karıGımı (Cu, Zn, Pb ve Fe) uygulamalarının etkilerinin moleküler düzeyde belirlenmesi amaçlandı. Bu amaçla hücrelere içme sularında izin verilen konsantrasyon ve 10 kat yüksek konsantrasyonda metal karışımı 24 ve 48 saat süre ile uygulandı ve antioksidan enzimler, ısı şok proteinleri ve apoptoz yolak genlerinin ifadeleri ile DNA polimorfizmi belirlendi. Çalışma sonucunda ağır metal karışımına maruz kalan hücrelerde DNA polimorfizminin arttığı, antioksidan enzim gen ifadelerinin ve apoptotik hücre oranı ile birlikte apoptoz yolak genlerinin ifadelerinde artışın olduğu saptandı. Sonuç olarak, içme sularında izin verilen limitlerde ağır metal karışımının aorta düz kas hücrelerinde oksidatif stres, genotoksisite ve apoptoza neden olduğu belirlendi. Vasküler düz kas hücrelerinde apoptozun arteroskleroz, hipertansiyon gibi hastalıkların patofizyolojisinde rol oynadığı göz önünde bulundurulduğunda bu limitlerin daha kapsamlı çalışmalarda araştırılarak değerlendirilmesi gerektiği düşünüldü.
Heavy metals are the most common pollutants with specific gravity greater than 5 g cm-3. Exposure of people to heavy metals is mostly caused by the consumption of food and water containing heavy metals. Metals taken with drinking water can cause genotoxicity and various diseases in the body. For this reason, the permissible heavy metal limits in water used for human consumption have been determined by laws and regulations. In our study, it was aimed to determine the effects of metal mixture (Cu, Zn, Pb and Fe) application at the limits allowed in drinking water of human healthy aorta smooth muscle cells (TG-HA-VSMC) at the molecular level. For this purpose, the mixture of metal in the drinking water and the ten fold higher concentration in the drinking water was applied for 24 and 48 hours and DNA polymorphism was detected with the expressions of antioxidant enzymes, heat shock proteins and apoptosis pathway genes. At the end of the research it was stated that the expression of DNA polymorphism, antioxidant enzyme gene expression and apoptotic cell ratio as well as apoptosis pathway genes were increased in cells exposed to heavy metal mixture. As a result, it was determined that heavy metal mixture caused oxidative stress, genotoxicity and apoptosis in aorta smooth muscle cells at the limits allowed in drinking water. Considering that it played a role in the pathophysiology of diseases such as apoptosis arthrosclerosis and hypertension in vascular smooth muscle cells, it was thought that these limits should be investigated and evaluated in more comprehensive studies.
