Özet
Bu çalışmada gıda endüstrisinde ve evlerde ıspanağın temizlenmesiyle açığa çıkan değerli bir atık olan ıspanak kökünden geleneksel ekstraksiyon yöntemlerine alternatif olarak yeşil ve yenilikçi yöntemler arasında özellikle uygulama süresinin kısa olması nedeniyle dikkat çeken ultrason destekli ekstraksiyon yöntemiyle biyoaktif bileşenlerin kazanılması hedeflenmiştir. Ultrason destekli ekstraksiyon prob tipi ultrasonik homojenizatör ile gerçekleştirilmiş ve yöntemde ekstraksiyon değişkenleri genlik (%25-55), sıcaklık (10-30 °C), süre (3-5 dk) ve etanol konsantrasyonu (%20-60) olarak belirlenmiştir. Optimizasyon sürecinde ekstraksiyon değişkenlerinin ıspanak kökünün toplam fenolik madde miktarı, toplam flavonoid madde miktarı ve antioksidan aktivitesi üzerine olan etkileri incelenmiştir. Süreç, yanıt yüzey yöntemi ve merkezi kompozit tasarım kullanılarak 30 farklı deneme koşulu içeren bir deney tasarımı olacak şekilde tasarlanmış ve bu koşullar için optimize edilmiştir. Ispanak kökünde maksimum miktarda fenolik bileşiğin (5579,51 mg GAE/kg LF), flavonoid maddenin (1824,34 mg KE/kg LF) ve antioksidan aktivitenin (20,79 mmol TE/kg LF) elde edildiği ekstraksiyon koşulları %55 genlik, %20 etanol konsantrasyonu, 10 °C sıcaklık ve 3 dk ekstraksiyon süresi olarak belirlenmiştir. Optimum ekstraksiyon koşulunda elde edilen ıspanak kökü ekstraktının toplam fenolik madde miktarının %31,55 oranında ve toplam flavonoid madde miktarının %66,80 oranında arttığı, antioksidan aktivitesinin ise %11,15 oranında azaldığı tespit edilmiştir.
In this study, it was aimed to obtain bioactive components from spinach root, which is a valuable waste released by cleaning spinach in the food industry and homes, by ultrasound-assisted extraction method, which is among the green and innovative methods as an alternative to traditional extraction methods and attracts attention especially due to the short application time. Ultrasound assisted extraction was performed with probe type ultrasonic homogenizer and extraction variables were determined as amplitude (25-55%), temperature (10-30 °C), time (3-5 min) and ethanol concentration (20-60%) in the method. The effects of extraction variables on the total phenolic substance amount, total flavonoid substance amount and antioxidant activity of spinach root were investigated in the optimization process. The process was designed to be an experimental design with 30 different experimental conditions using the response surface method and a central composite design and optimized for these conditions. Extraction conditions in which the maximum amount of phenolic compounds (5579.51 mg GAE/kg LF), flavonoid substance (1824.34 mg KE/kg LF) and antioxidant activity (20.79 mmol TE/kg LF) were extracted from the spinach root, were determined as 55% amplitude, 20% ethanol concentration, 10 °C temperature and 3 min extraction time. It was found that the total amount of phenolic substance and the total amount of flavonoid substance of spinach root extract obtained under optimal extraction conditions increased by 31.55% and 66.80% respectively, while the antioxidant activity decreased by 11.15%.
