I(Yüksek Lisans Tezi)?Gramineae Familyasına ait Bazı Türlerin Polen Morfolojisinin İncelenmesi vePolen Ekstrelerinin Hazırlanması?Trakya ÜniversitesiFen Bilimleri EnstitüsüBiyoloji Anabilim DalıÖZETBu çalışmada, Trakya bölgesinde üretimi yapılan, Gramineae familyasına ait 5türün (Avena sativa L., Triticum aestivum L., Secale cereale L., Hordeum vulgare L.,Oryza sativa L.) alerjen polenlerinin morfolojileri ışık ve elektron mikroskobu ileincelenmiş ve bu türlerden polen ekstreleri hazırlanmıştır.Morfolojik çalışmalarda kullanılan materyaller, Trakya Tarımsal AraştırmaEnstitüsünden temin edilmiştir. Polen preparatları, Wodehouse (1935) metoduylahazırlanmış, polenlerin morfolojik özellikleri ışık ve elektron mikroskobundasaptanmış, daha sonra mikrofotografları çekilmiştir.Işık mikroskobu ile yapılan morfolojik çalışmalar sonucunda, polenboyutlarının, türler arasında farklılıklar gösterdiği ve polenlerin ortak özellikleri olaraksphaeroidea, oblata-sphaeroidea, monoporatae, operculate ve ekzin'in tectatae olduğubelirlenmiştir.SEM incelemelerine göre Avena sativa L., Triticum aestivum L., Secale cerealeL. ve Hordeum vulgare L. için ekzin skülptürleri spinulose iken Oryza sativa L.'dagranüle olduğu görülmüştür.Toplanan polenlerin ekstrelerinin hazırlanması için Aytuğ ve Arkadaşları (1991)tarafından uygulanan ekstraksiyon yöntemi kullanılmış, ekstraktif olarak Cocasolüsyonu ve sterilizasyon için steril filtrasyon tekniği uygulanmıştır.Elde edilen ham polen ekstreleri bu bölge florasında yetişmiş olan bitkilereaittir. Alerjik hastalıkların teşhisinde yararlanabileceğimiz bu ham ekstraktlar birçokmajor ve minör alerjenler içermektedir. Pollen preparations have been made throughWodehouse (1935) method, morphological features of the pollen grains have beendetermined through light and electron microscopes and then photomicrographs ofpollens were taken.As a result of the morphological studies of pollens with light microscope, it hasbeen understood that pollens? size showed differences among species and as commonfeatures of pollens it were determined sphaeroidea, oblata-sphaeroidea, monoporatae,operculate and exine were tectatae.According to SEM examinations while exine sculptures were spinulose forAvena sativa L., Triticum vulgare L., Secale cereale L. and Hordeum vulgare L,granule was observed in Oryza sativa L.The extraction method which was applied by Aytuğ et. al. (1991) was used forpreparing the extracts of collected polen, as an extractive Coca solution and forsterilization sterile filtration technique were used.The crude polen extracts which were obtained belongs to plants grown in thisregion. These crude extracts ,we can use them for diagnosis allergic diseases, includelots of major and minor allergens.
