Özet
Luteinleştirici hormon (LH) ön-hipofiz bezinden salgılanır ve üreme sisteminin gelişimi ve fonksiyonunda önemli rol oynar. LH cinsiyet bezlerinin normal fonksiyonu ve üreme için gereklidir. Erkeklerde testosteron üretimini uyarırken, kadınlarda östrojen ve progesteron üretimini uyarır ve yumurtlamayı başlatır. Kadınlarda artan LH seviyeleri primer gonadol bozukluk, polikistik over sendromu, post-menapoz ve hipofiz adenomları ile ilişkilendirilmektedir. Erkeklerde ise hipogonadizm ve primer testis bozukluğunda yüksek LH konsantrasyonu görülür. Bu tezde; elektrokimyasal impedans spektroskopisi (EIS) ile LH'ın kantitatif tayini için spesifik ve duyarlı immünolojik bir biyosensör geliştirildi. 12-merkaptododekanoik asit, EDC/NHS çifti, PAMAM ve glutaraldehit kullanılarak kovalent immobilizasyon yöntemi ile anti-LH antikoru altın elektrot yüzeyine immobilize edildi. İmmobilizasyon adımları siklik voltammetri ve elektrokimyasal impedans spektroskopisi ile karakterize edildi. Bu biyosensörün tayin süresi 1 saat olup, biyosensörün tayin aralığı 1-60 mIU/mL LH konsantrasyonu aralığı olarak belirlendi. Ayrıca geliştirilen biyosensör, LH eklenmiş yapay serum örneklerinin analizinde de başarıyla kullanıldı.

Luteinizing hormone (LH) is secreted by the anterior pituitary gland, and plays a key role in the development and function of the reproductive system. LH is essential for normal gonadal function and fertility, stimulating testosterone production in men while it stimulates estrogen and progesterone production and triggers ovulation in females. Increased LH levels are related with primary gonadal dysfunction, polycystic ovary syndrome, post-menopause and pituitary adenoma in females. High LH concentration has been found in men with hypogonadism, and primary testicular failure. In this thesis, a specific and sensitive immunological biosensor was developed for quantitative detection of LH by electrochemical impedance spectroscopy. Anti-LH antibody was immobilized onto gold electrode surface via covalent immobilization techniques by using mercaptododecanoic acid, EDC/NHS couple, PAMAM and glutaraldehyde. Immobilization steps were characterized by cyclic voltammetry and electrochemical impedance spectroscopy. The detection time of this developed biosensor system was one hour. Its detection range was determined as 1-60 mIU/mL LH concentration during this period. This constructed biosensor was successfully used to artificial serum samples spiked with LH.
