Abstract
Bu çalışmada, Helianthus annuus L. (ayçiçeği) bitkisinden embriyo kültürü yöntemi kullanılarak elde edilmiş olan bitkilerin soğukta muhafaza edilerek korunmasına ve bu bitkilerde oluşan anatomik ve karyolojik değişimlerin belirlenmesine çalışılmıştır. Çalışma materyali olarak kullanılan ayçiçeği tohumları Trakya Tarımsal Araştırma Enstitüsü'nden sağlanmıştır. Çalışmalar, Trakya Üniversitesi Fen-Edebiyat Fakültesi Biyoloji Bölümü Doku Kültürü Laboratuvarı'nda yapılmıştır. Çalışma, deney ve kontrol grubu olacak şekilde yapılmıştır. Kontrol grubu bitkileri kum ortamında çimlendirilerek elde edilmiştir. Deney grubu bitkileri ise, MS besi ortamında embriyo kültür yöntemiyle yetiştirilen bitkiler ve bunların bir kısmının düşük ısıda muhafaza edilmesiyle elde edilen bitkilerden oluşmuştur. Yapılan anatomik değerlendirmede, deney grubu bitkilerinin epiderma, korteks parankiması, parankima, iletim demetleri gibi yapılarının kontrol grubuna göre farklılıklar gösterdiği belirlenmiştir. Mitotik indeks değerleri kontrol grubunda %22.98, deney grubu kültür ortamında yetişen bitkilerde %60.00, soğukta muhafaza edilmiş bitkilerde ise %10.15 olarak belirlenmiştir. In this study, plants which were obtained by applying embryo culture method to the Helianthus annuus L. plants, were tried to protect in cold and tried to determine anatomical and caryological changes
in the plants. The sunflower seeds that were used in work, were supplied from Trakya Agriculture Research Institue. All works were done in Trakya University Faculty of Art and Science Biology Department Tissue Culture Laboratory. The work was performed in two groups: Experiment and Control. Control group was obtained by turfing plants in the sand medium. On the other hand experiment group was consisted of plants that were grown by the method of embryo culture in MS medium and plants which were grown up either at the low temperature and by the method of embryo culture. In the anatomical discussion, it is seen that, plant parts such as epidermis, cortex parenchyma, parenchyma, vascular bundles show disparities in the control and experiment group. Also mitotic index value was determined : %22.98 in the control group plants, %60.00 in the experiment group plants and %10.15 in the plants that were grown in cold.
