BitkisindekiFenolik Bileşiklerin zolasyonu ve TanımlanmasıTrakya ÜniversitesiFen Bilimleri EnstitüsüKimya Ana Bilim DalıÖZETBu tez çalışmasında Hypericum montbretii bitkisinin kimyasal bakımdanincelenmesi amaçlanmıştır. Bu amaçla metanol içinde 2 hafta bekletilen bitkinin ekstresisırası ile hekzan, kloroform, etilasetat ve butanol ile ekstrakte edilmiştir. Elde edilen bufraksiyonların çift boyutlu kağıt kromatografileri yapılmış ve etilasetat ve butanolfraksiyonlarının fenolik bileşikleri içerdiğine karar verilmiştir. Maddelerin bufraksiyonlardan eldesinde çeşitli kromatografik yöntemlerden yararlanılmıştır.Toplam 12 bileşik izole edilmiştir. Bileşiklerden yedisi flavonoid, üçü fenilpropanoid, biri ksanton, biri de şikimik asittir.Flavonoid bileşikleri, kersetin, kamferol, I 3 II 8′′ biapigenin, kersitrin, hiperin,mirisitrin ve rutindir. Fenil propanoid bileşikleri metil klorojenat, klorojenik asit ve 3-O-p-kumaroilkuinik asittir. Ksanton bileşiği ise bir C-glikozil ksanton olan mangiferindir.Bileşiklerin tümü yeni olmayıp daha önce Hypericum türlerinden ve diğer bazı bitkitürlerinden izole edilmişlerdir.Bileşiklerin yapı tayininde IR, UV, 1H-NMR, 13C-NMR, DEPT, HMBC, COSY,FAB MS, EI MS gibi spektroskopik tekniklerden yararlanılmıştır. Ayrıca standart maddeile TLC'de karşılaştırma yapılarak ve literatür çalışmaları ile kesin sonuca varılmıştır.Bulgular ayrıntılı olarak açıklanmıştır.Yıl: 2005, 165 sayfa.Anahtar kelimeler: Hypericum montbretii, flavonoid glikozidler, ksanton C-glikozid,klorojenik asid ve 3-O-p-kumaroil kuinik asid.

Forthis purpose, the plant material soaked in MeOH for two weeks. Then MeOH extract wasfractionated with hexane, chloroform, ethyl acetate and butanol, respectively.Thesefractions were checked by two dimensional paper chromatography. From the results ofpaper chromatography, it was decided that ethylacetate and butanol fractions containphenolic compounds. Several chromatographic techniques have been used for the isolationof the compounds from the extracts.Totally 12 compounds were obtained. Seven of them were flavonoids, three of themwere phenylpropanoids, one of them was xanthone glycoside and one of them was shicimicacid.The flavonoid compounds were quercetin, kaempferol, I 3 II 8′′ biapigenin,quercitrin, hyperin, myricitrin and rutin. Phenylpropanoids were methylchlorogenate,chlorogenicacid and 3-O-p-coumaroyl quinicacid. The xanthone compound was aC-glycoside xanthone, mangiferin.All of them were known compounds. The structure determination of the compoundsachieved by spectral methods such as IR, UV, 1H-NMR, 13C-NMR, HMBC, COSY, FABMS and EI MS and also by comparing their spectral data to those of literature values and byTLC comparison with authentic samples.The results were discussed.Year: 2005, 165 pages.Keywords: Hypericum montbretii, flavonoid glycosides, xanthone C-glycoside, chlorogenicacid and 3-0-p-coumaroyl quinicacid.
