Özet
Kanser biyobelirteçleri, onkolojik rahatsızlıklarda bulundukları dokuda yüksek miktarda üretilirler ve kana yüksek miktarlarda karışırlar. Biyobelirteçlerin kandaki miktarlarının tespiti kanserin erken teşhisi ve tedavisinin izlenmesinde önemli rol oynar. Trombospondin-1 (THBS1 veya TSP1) hücre-hücre ve matriks-hücre etkileşimlerini yönlendiren bir glikoproteindir. Literatürde pek çok kanser türüyle ilişkilendirilse de, Uluslararası Kanser Enstitüsü (NCI)’nün Erken Tanı Araştırma Ağı’nda (EDRN) belirtildiği üzere prostat kanserinin erken tanısında daha belirgin özellikler göstermektedir. THBS1’in kanser biyobelirteci olarak kabulü ile ilgili devam eden çalışmalarda faz-2 klinik deneme aşamasına gelinmiştir. Bu tez kapsamında, elektrokimyasal impedans spekroskopisine (EIS) dayalı Trombospondin biyosensörü geliştirildi. Trombospondin-1’e spesifik antikor (Anti-THBS1), altın elektrot yüzeyine 6-merkaptohekzanol, epiklorohidrin, etanolamin kullanılarak kovalent olarak immobilize edildi. Hazırlanan biyosensör 50-300 ng/mL tayin aralığında ve 1 saat tayin süresinde THBS1 tayininde başarıyla kullanıldı.

Cancer biomarkers are mixed with blood by being produced in large amounts in the tissue during the oncological diseases. The determination of their amounts in the blood plays important role in early diagnosis and treatment monitoring of cancer. Thrombospondin-1 (THBS1 or TSP1) is a glycoprotein that leads cell-cell and matrix-cell interactions. Even though it is associated with a lot of type cancer in literature, it indicates more typical characteristics in early diagnosis of prostate cancer, as mentioned in Early Detection Research Network (EDRN) of National Cancer Institute (NCI). The on-going studies interested in the consideration of THBS1 biosensor based on electrochemical impedance spectroscopy was developed. Anti-THBS1, which is specific antibody to THBS1, was covalently immobilized onto gold electrode surface by using 6-mercapto-1-hexanol, epichlorohydrin and ethanolamine. The constructed biosensor was successfully used in the detection range between 0-300 ng/mL and detection time of 1 hour for THBS1 analysis.
