Abstract
Bu çalışmanın amacı, Malvaceae familyasına ait Hibiscus sabdariffa L. bitkisinin antimikrobiyal ve antioksidan aktivitesinin çeşitli metotlarla incelenmesidir. Bu amaçla kurutulup öğütülen bitkilerin su, etanol ve aseton çözücüleri kullanılarak ekstraksiyonları yapıldı. En yüksek ekstraksiyon verimi su ekstraktında gözlendi. Bütün ekstraktların toplam fenolik madde içeriği Folin-Ciocalteu ayıracı ile belirlendi. Onların toplam antioksidan aktiviteleri DPPH serbest radikali giderme metodu, indirgeme kapasitesi metodu ve ferrik tiyosiyanat (FTC) metodu kullanılarak tayin edildi. Elde edilen sonuçlar ?-tokoferol, BHT ve BHA standart maddeleriyle kıyaslanarak değerlendirildi. Ekstraktların antimikrobiyal aktiviteleri disk difüzyon yöntemi kullanılarak belirlendi. Bitki ekstraktlarının antimikrobiyal aktivitesinin belirlenmesi için 5 bakteri (Staphylococcus aureus ATCC 29213, Escherichia coli ATCC 25922, Pseudomonas aureginosa ATCC 27853, Klebsiella pneumoniae ATCC 700603, Enterococcus faccialis klinik izolat) ve 2 maya (Candida albicans ATCC 60193, Candida crusei 6258) türü kullanıldı. Toplam fenolik madde tayini sonucunda, ekstraktların toplam fenolik madde miktarlarının gallik asit eşdeğeri olarak 53.84±0,008 – 45.38±0.002 mg/g aralığında değiştiği belirlendi. Etanol ekstraktı en yüksek fenolik madde miktarına sahipti. Serbest radikal giderme aktivitesinden elde edilen verilere göre su ve etanol ekstraktlarının 500 ve 1000 µg/mL’lik konsantrasyonları standart maddelerle karşılaştırılabilir düzeyde DPPH giderme aktivitesi gösterdi. Aseton ekstraktının DPPH giderme aktivitesi bakımından zayıf olduğu gözlendi. ii Bitki ekstraktlarının toplam antioksidan aktivitesi linoleik asit emülsiyonunda değerlendirildi. Elde edilen sonuçlara göre su, etanol ve aseton ekstraktlarının 50 ve 100 µg/mL konsantrasyonları haricindeki tüm ekstraktların etkili ve özellikle 500 ve 1000 µg/mL konsantrasyonlarda yüksek oranlarda antioksidan aktivite gösterdiği belirlendi. İndirgeme kapasitesi tayini sonuçlarına göre bütün bitki ekstraktlarının standartlarla kıyaslandığında yüksek aktiviteye sahip olmadığı gözlendi. H.sabdariffa‘dan hazırlanan ekstraktlar arasında aseton ekstraktının en yüksek antimikrobiyal aktiviteyi gösterdiği ve en duyarlı olduğu bakterinin S. aureus olduğu tespit edildi.
Abstract
The aim of this study was to investigate antimicrobial and antioxidant activities of Hibiscus sabdariffa belonging to Malvecea Family. For this purpose, the extractions of dried and milled plants were carried out by using water, ethanol and acetone as solvent. It was observed the highest extraction yield in the water extract. The total phenolic compound contents of all extracts were determined by Folin- Ciocalteu reagent. Their total antioxidant activities were assayed by using DPPH free radical scavenging method, recoding power method and ferric thiocyanate method. The obtained results were evaluated by comparing to ?- tocopherol, BHT and BHA as standard. Antimicrobial activities of extracts were determined by disc diffusion method. For that purpose 5 species of bacteria (Staphylococcus aureus ATCC 29213, Escherichia coli ATCC 25922, Pseudomonas aureginosa ATCC 27853, Klebsiella pneumoniae ATCC 700603, Enterococcus faccialis clinical isolate) and 2 species of yeast(Candida albicans ATCC 60193, Candida crusei 6258) were used. At the end of total phenolic compound assay the phenolic contents were determined to be in the range 53.84±0.008 – 45.38±0.002 mg/g as gallic acid equivalent. Ethanol extract had the highest phenolic content. According to the free radical scavenging activity, water and ethanol extract showed DPPH scavenging activity which is comparable with standard compound at the 500 and 1000 mg/ mL concentration. Acetone extract showed weakly DPPH activity. Total antioxidant activities of the plant extracts were assayed by using linoleic acid emulsion. According to obtained data all extracts showed effective antioxidant activity except for 50 and 100 mg/ml concentrations. Especially their antioxidant activities were very high at 500 and 1000 mg/ml concentrations. All extracts hadn’t high reducing capacity as compared to standards. Among the prepared extracts from H. sabdariffa acetone extract showed the highest antibacterial activity and S.aureus were determined to be most sensitive bacterium
