Özet
Bu çalışma kapsamında alabaş (Brassica oleracea var. Gongylodes) bitkisinin antioksidan aktivitesi çeşitli metodlarla incelenmiştir. Bu amaçla bitkinin taze yaprak ve gövdesinin ayrı ayrı, etanol, metanol, aseton ve su çözücüleri kullanılarak ekstraksiyonları yapıldı. Her bir ekstraktın Folin-Ciocalteu ayıracı (FCR) ile toplam fenolik madde içeriği, DPPH serbest radikali giderme aktivitesi, indirgeme gücü, metal iyonlarını şelatlama kapasitesi, süperoksit radikali giderme aktivitesi ve ß-Karoten ağartma yöntemi kullanılarak antioksidan aktivitesi tayin edildi. Elde edilen sonuçlar standart madde olarak kullanılan C vitamini, E vitamini, BHA ve BHT ile karşılaştırılarak değerlendirildi. Etanol, metanol, aseton ve su ile yapılan ekstraksiyonlar sonunda bitkinin kurutulmuş yaprak ve gövdesinden ekstrakte edilebilen maddelerin yüzde verimi; sırasıyla % 3,05-% 8,619 arasında bulunmuştur. Yaprak ve gövde ekstraktlarının fenolik madde içerikleri toplam fenolik madde miktarlarının gallik asit eşdeğeri olarak hesaplandı. En yüksek fenolik madde içeriği yaprağın aseton ekstraktında tayin edildi. DPPH radikali giderme aktivitesinin tayin sonucları; gövde ekstraktları arasında en iyi aktiviteye metanol ekstraktının, yaprak ekstraktları arasında ise aseton ve etanol ekstraktlarının sahip olduğunu göstermiştir. Standart maddelere yakın aktivite gösteren ekstraktlar; yaprağın aseton, etanol ve metanol ekstraktlarıdır. Metal şelatlama aktivitesi deneyinde, standartlara en yakın aktivite gösteren ekstrakt yaprağın su ekstraktıdır. Yaprak ve gövde kısımları kendi aralarında kıyaslandıklarında ise her ikisinde de en iyi aktiviteyi su ekstraktları göstermiştir. ß-Karoten ağartma yöntemi kullanılarak yapılan antioksidan aktivite tayininde gövde ekstraktları aktivite göstermezken, yaprak ekstraktları aktivite göstermiştir. En yüksek aktivite yaprağın su ekstraktında gözlenmiştir. Süperoksit radikali giderme aktivitesi tayininde; elde edilen verilere göre yaprak ve gövde örneği ayrı ayrı incelendiğinde her ikisinde de en yüksek aktiviteyi su ekstraktlarının gösterdiği gözlenmiştir. Ekstraktların indirgeme gücü tayininde; ekstraktların aktiviteleri standartlarla karşılaştırıldığında düşük aktiviteye sahip oldukları gözlendi. Kendi aralarında incelendiğinde yaprağın aktivitesinin gövdeden yüksek olduğu belirlendi.
In this study, antioxidant activities of kohlrabi (Brassica oleracea var. Gongylodes) were investigated by using different methods. For this purpose, the extractions were done from leaves and stem of the fresh plant by using water, ethanol and acetone as solvent. The antioxidant activities of all extracts were assayed with the various methods including total phenolic compound contents by Folin-Ciocalteu reagent (FCR), DPPH free radical scavenging activity, reducing capacity, metal chelating capacity, superoxide anion scavenging activity, and ß-caroten bleaching test. The obtained results were compared by using the vitamins C and E, BHT and BHA as standard materials. Percentage yield of extractable compound of all studied plants were found in the range between 3,05 % and 8,619 % mg/g from dried leaves and stem of kohlrabi at the end of the extractions carried out by water, ethanol, methanol and acetone as solvent. Total phenolic compound amounts of leave and stem extracts were determined as gallic acid equivalent. The highest phenolic compoundamounts were found in the acetone extract of the leave. The assay result of DPPH radical scavenging activity showed methanol extract had the best activity among the stem extracts; and acetone and ethanol extracts had the best activity among the leave extracts. The extracts which showed an activity nearly the standard materials were the acetone and ethanol extracts of the leave. In metal chelating activity experiment, the water extract of the leaves showed an activity near the standards. When compared in each other, the water extracts of leaves and stem showed the best activity. In the assay of total antioxidant activity done with ß-carotene bleaching test, the stem extracts didn?t show any activity but leave extracts showed. The best activity was observed in the water extract of leaves. According to the data for the measurement of superoxide anion scavenging activity, the water extracts of leave and stem showed the best activity. When the reducing power assays of the extracts were examined, extracts, when compared with the standards, were observed low activity. Leave activity was higher than the stem activity.
