Özet
Bu çalışmada, birçok gıdamızda renklendirici olarak kullanılan ve Brilliant blue (E133) olarak bilinen mavi gıda boyası, laboratuvar ortamında Drosophila .melanogaster besiyerlerine eklenerek, boyanın bu sineklerdeki sitotoksik ve genotoksik etkisi araştırılmıştır. Çalışmanın ilk kısmında, mavi boya yüksek dozda (2gr/Lt) standart eklenmiştir. Bu besiyerlerde yetiştirilen larva, pupa ve ergin sayıları kontrol grubu (Distile su) ile karşılaştırılmıştır. Yavru sayıları karşılaştırılan gruplarda istatistiki testler anlamlı bir sonuç vermediğinden (P >0.05) mavi boyanın yavru veriminde herhangi negatif bir etki göstermediği görüşüne varılmıştır. Çalışmanın ikinci kısmında, mavi boyanın Drosophila melanogaster’deki genotoksik etkisi SMART (Somatik Mutasyon ve Rekombinasyon Testi) yöntemi kullanılarak araştırılmıştır. Larva aşamasında yoğun mavi boya ile muamele edilen besiyerlerden elde edilen ergin sineklerin kanatlarında mutasyon sonucu ortaya çıkan anormal beneklenmelerin sayıları, kontrol grubundaki sineklerin kanatlarındakilerden oldukça fazla olması (P<0.001), mavi boyanın son derece mutagenik olabildiğini göstermiştir.
In this study, the brilliant blue (E133) which is commonly used in the food industry as a colouring additive is examined for its cytotoxic and genotoxic effects using Drosophila melonagaster in the laboratory conditions. In the first part of the work, a high concentration (2gr/Lt) of the blue coluring agent was applied to mediums where Drosophila melanogaster flies were reared in breeding bottles. The numbers of the larvae, pupae and the adult flies were compared to control (distilled water) groups to see the cytotoxic effects. However, the statistical tests revealed no significant difference (P > 0.05) among the fly numbers in appropriate groups. Thus, it seemed that blue food colouring agent has no decreasing effect on the fly numbers. The second part of the work, the genotoxic effects of the blue colouring agent was examined using SMART (Somatic Mutation and Rekombination Test) technique on Drosophila melanogaster. The larvae were chronically fed in the mediums where blue colouring agent were experimentally applied. The results showed that this food additive has highly significant effects (P < 0.001) for inducing the number of mutant wing spots compared to control groups. Thus, it seemed that the blue food colouring is a highly mutagenic agent on Drosophila melanogaster offspring in the laboratory conditions.
