Öğe The effect of high protein diet and exercise on irisin, eNOS, and iNOS expressions in kidney(Taylor & Francis Ltd, 2016) Tastekin, Ebru; Palabiyik, Orkide; Ulucam, Enis; Uzgur, Selda; Karaca, Aziz; Vardar, Selma Arzu; Yilmaz, AliLong-term effects of high protein diets (HPDs) on kidneys are still not sufficiently studied. Irisin which increases oxygen consumption and thermogenesis in white fat cells was shown in skeletal muscles and many tissues. Nitric oxide synthases (NOS) are a family of enzymes catalyzing the production of nitric oxide (NO) from L-arginine. We aimed to investigate the effects of HPD, irisin and NO expression in kidney and relation of them with exercise and among themselves. Animals were grouped as control, exercise, HPD and exercise combined with HPD (exercise-HPD). Rats were kept on a HPD for 5 weeks and an exercise program was given them as 5 exercise and 2 rest days per week exercising on a treadmill with increasing speed and angle. In our study, while HPD group had similar total antioxidant capacity (TAC) levels with control group, exercise and exercise-HPD groups had lower levels (p<0.05). Kidneys of exercising rats had no change in irisin or eNOS expression but their iNOS expression had increased (p<0.001). HPD-E group has not been observed to cause kidney damage and not have a significant effect on rat kidney irisin, eNOS, or iNOS expression. Localization of irisin, eNOS, and iNOS staining in kidney is highly selective and quite clear in this study. Effects of exercise and HPD on kidney should be evaluated with different exercise protocols and contents of the diet. risin, eNOS, and iNOS staining localizations should be supported with various research studies.Öğe The effect of L-lysine on serum sialic acid levels in isoproterenol-induced myocardial infarction in rats(Turkish Biochem Soc, 2011) Uzgur, Selda; Usta, Ufuk; Goekmen, Selma SueerObjectives: To investigate the effect of L-lysine on serum total and lipid-bound sialic acid levels in isoproterenol-induced myocardial infarction and to evaluate the role of cell damage in the elevation of sialic acid post-infarction. Material and Methods: Male albino rats of Wistar strain were divided into three groups randomly: control, isoproterenol and isoproterenol+L-lysine. Myocardial infarction was produced with 150 mg/kg of isoproterenol administered intraperitoneally twice at an interval of 24 hour. L-Lysine was given orally (5 mg/kg/day) for 5 days. Existence of experimental infarction was confirmed by histopathological changes and the elevation of troponin I. The levels of serum total and lipid-bound sialic acid were determined by the methods of Warren and Katopodis, respectively. Results: Isoproterenol caused a significant elevation in serum troponin I, total and lipid-bound sialic acid levels and a prominent atrophy and fibrotic changes confirming myocardial infarction in heart tissue. L-Lysine significantly prevented troponin I, total and lipid-bound sialic acid increase in serum and atrophy and connective tissue development in heart tissue. Conclusion: Cell damage may play an important role in serum sialic acid elevation after myocardial infarction. Conflict of interest: The authors have no conflict of interest.Öğe L-arginin ve L-lizinin deneysel ateroskleroz gelişimine etkisinin incelenmesi(Trakya Üniversitesi Sağlık Bilimleri Enstitüsü, 2022) Uzgur, Selda; Süer Gökmen, SelmaBu çalışmanın amacı; L-arginin ve/veya L-lizinin aterosklerozun önlenmesindeki rolünü ortaya koymaktır. 80 adet Wistar albino erkek sıçan; tedavi verilmeyen, lizin verilen, arginin verilen ve lizin+arginin verilen kontrol ve ateroskleroz grupları olmak üzere eşit sayıda rastgele 8 gruba ayrıldı. Deneysel ateroskleroz modeli oluşturmak için hayvanlara 12 hafta süresince %3 kolesterol ve %1 kolik asit içeren diyet uygulandı. Arginin %3 oranında, lizin 12 mg/kg olacak şekilde sıçanların içme suyuna katılarak 12 hafta boyunca verildi. Ateroskleroz modeli, lipid-lipoprotein düzeyleri ölçülerek ve histopatolojik olarak ortaya konuldu. Serum trigliserit, total kolesterol ve HDL kolesterol düzeyleri otoanalizörde kit kullanılarak ölçüldü. Serum arginaz aktivitesi Corraliza metodu, serum ornitin düzeyleri Chinard metodu, serum ADMA/arginin düzeyleri Teerlink metodu ve NO düzeyleri Cortas metodu ile ölçüldü. Serum TAS ve TOS, Rel Assay Diagnostic kiti kullanılarak ELISA yöntemi ile ölçüldü. OSİ, aterojenik indeks, LDL ve VLDL kolesterol düzeyleri formül kullanılarak hesaplandı. Lizin ve/veya arginin tedavisi deneysel aterosklerozda gözlenen lipid düzensizliğini, oksidatif stresi, arginaz aktivitesindeki ve ADMA düzeylerindeki artışı önledi. Sonuç olarak, sıçanlarda tek başına ya da birlikte verilen lizin ve argininin aterosklerozun önlenmesinde etkili olduğu bulundu.
