Özet
ÖZET Sklerozan kolanjit günümüzde önemli bir sağlık sorunudur. Medikal ve cerrahi tedavisi yapılan bütün çalışmalara rağmen yüz güldürücü değildir. Bu nedenle deneysel hayvan modelinde medikal tedavide kullanılabilecek melatoninin bir skolosidal ajan olan formolle oluşturulan sklerozan kolanjit üzerine etkilerini araştırdık. Çalışmada, Trakya Üniversitesi Tıp Fakültesi Deney Hayvanları Araştırma ve Uygulama Biriminden temin edilen Wistar-Albino soyu, sağlıklı, 190-220 gram ağırlığında, 40 adet, 3,5-4 aylık, genç, erkek sıçanlar kullanıldı. Sıçanlar; oda ısısı 21±2°C de sabit sıcaklıkta, %50 nem oranında, 12 saat aydınlıkta ve 12 saat karanlıkta tutuldular. Ratlar 4 grup halinde çalışmaya alındı, l.grup kontrol, 2. grup skolosidal ajan (formol), 3. grup melatonin, 4. grup formol + melatonin grubu olarak ayrıldı. 2 ve 4. gruba koledok içine skolosidal ajan (formol), 3 ve 4. gruba intraperitoneal olarak melatonin, 1. gruba ise koledok içine izotonik verildi. Ratlara 60 gün sonra karaciğer ve safra yolları sintigrafisi çekildi. Sintigrafiden sonra sakrifiye edilen hayvanlardan ekstrahepatik safra yolları ve karaciğer, biopsisi alınarak histolojik incelemeye gönderildi. Deneysel olarak oluşturulan sklerozan kolanjit modelinde melatoninin histopatolojik bulguların gelişmesini engellediği görülmüştür. 45
THE EFFECTS OF MELATONIN ON EXPERIMENTAL SCLEROSING CHOLANGITIS IN RATS SUMMARY Nowadays sclerosing cholangitis is a serious health problem. There is not a hopeful medical or surgical treatment though all recent researchs. Therefore, we investigated the effects of melatonin on the sclerosing cholangitis induced by a scolosidal agent formaline. In this study, we used 199-220 grams, healthy, 3-4 months old, and male Wistar-Albino rats taken from Trakya University Medical Faculty Experimental Research Unit. The rats were sheltered in the dark and the daylight for 12 hours where there was 50% of moisture and 21 ± 2 ° C of temperature. The rats were divided into 4 groups; as isotonic saline (Group 1), formaline (Group 2), melatonin (Group 3) and melatonin+formaline (Group 4). Formaline was injected into the choledoc of Group 2 and 4, isotonic saline was injected into the choledoc of Group 1, and melatonin (10 mg/kg) was injected to group 3 and 4 intraperitoneally. The hepatobiliary scintigraphic investigation was made after 60 days. Extrahepatic bile duct and liver biopsies were taken for histopathological investigation after the sacrification of rats. We found that melatonin prevents the development of histopathological findings of experimental sclerosing cholangitis. 46
