Labada ( rumex patientia L.) ekstraktlarının İN VİTRO yöntemlerle antioksidan aktivitesinin ve fitokimyasal profilinin değerlendirilmesi

Özet
Labada (Rumex patientia L.) yaprağının taze ve kurutulmuş örneklerinin metanol ve su ekstraktları hazırlanarak serbest radikalleri giderme ve antioksidan aktiviteleri farklı metotlarla incelendi. En yüksek toplam fenolik bileşik, toplam flavonoid içeriği ve Demir (II) iyonlarını şelatlama aktivitesini taze labada yaprağının su ekstraktı göstermiştir. DPPH• radikali giderme akvitesinin tayininde, standartlara en yakın aktiviteyi taze labada yaprağının su ekstraktı (28.90±0,67) göstermiştir. H2 O2 giderme aktivitesinin tayininde, en yüksek H2 O2 giderme oranı taze labada yaprağının su ekstraktında belirlendi. Toplam ferrik (Fe +3 ) iyonlarını indirgeme kapasitesi tayininde, labada ekstraktlarının indirgeme kapasiteleri arasında taze-metanol>kuru-metanol>taze-su>kuru-su sıralaması gözlemlendi. Süperoksit anyon radikali giderme aktivitesi tayininde, en yüksek radikal giderme aktivitesini taze labada yaprağının su ekstraktı gösterdi. ABTS• + radikali giderme aktivitesinin tayininde, kuru labada yaprağının su ekstraktı (60.40±0,68) diğer labada yaprağı ekstraktlarından daha yüksek oranda radikal giderme aktivitesi gösterdi. Fosfomolibden metodu ile toplam antioksidan aktivite tayininde, taze labada yaprağının su ekstraktının daha yüksek aktiviteye sahip olduğu anlaşıldı. Klorofil içeriğinin belirlenmesinde, toplam klorofil miktarı (17.49 mg/kg) yaş ağırlık cinsinden incelendi. Karotenoid içeriğinin tayininde, kuru ağırlığın mg’ı başına düşen % toplam karotenoid içeriği (%10.82) tespit edildi. Hidroksil radikali giderme aktivitesini en yüksek taze labada yaprağının su ekstraktı gösterdi. Bakır iyonlarını indirgeme potansiyelinin tayininde, taze labada yaprağının su ekstraktı en yüksek indirgeme potansiyelini gösterdi. Linoleik asit sisteminde ferrik tiyosiyanat (FTC) metodu ile toplam antioksidan aktivite tayininde, en yüksek aktiviteyi kuru labada yaprağı metanol ekstraktının (83.81±0,50) gösterdiği tespit edildi. LC-MS/MS ile fenolik madde analizinde, en yüksek konsantrasyonun rutinde (12087,0758 ?g/kg), en düşük konsantrasyonun ise salisilik asitte (17,2674 ?g/kg) olduğu gözlemlendi. Labada (Rumex patientia L.) bitkisinin taze ve kuru örneklerinden elde edilen metanol ve su ekstraktlarının antioksidan aktivite gösterdiği ve taze labada yaprağının sentetik antioksidanlara alternatif olabilecek doğal bir antioksidan kaynağı olabileceğinin sonucuna varıldı.
The antioxidant activities of fresh and dried samples of (Rumex patientia L.) plant were by preparing methanol and water extracts, free radicals were examined by different methods. The highest total phenolic compound, total flavonoid content and iron (II) ions chelating showed the water extracts of the fresh dock. In the determination of DPPH• radical scavenging activity, the nearest activity to the standards showed the water extract of the fresh dock. In the H2 O2 scavenging activity assay, the highest H2 O2 removal rate was determined in the fresh extract of water. In the determination of total ferric ions (Fe 3+ ) reduction capacity, fresh-methanol>drymethanol>fresh-water>dry-water sequence was observed among the reduction capacities of extracts in the dock. In the determination of superoxide anion radical scavenging activity, the highest radical scavenging activity showed fresh extracts of water. In the determination of ABTS•+ radical scavenging activity, the water extract of the dry dock showed a higher rate of radical removal than the extracts of the other dock. The total antioxidant activity was determined by the phosphomolybdenum method, it was found that fresh water extract had higher. In determining chlorophyll content, total chlorophyll content (17.49 mg/kg) was examined in terms of wet weight. Total carotenoid content per mg of dry weight (%10.82) was determined in determination of carotenoid content. In determining the hydroxyl radical scavenging activity showed the highest of fresh water extract. In the determination of the reduction potential of copper ions, the fresh water extract showed the highest reduction potential. Determination of the total antioxidant capacity by the ferric thiocyanate method in linoleic acid sytem, the highest activity was determined by the methanol extract (%83.81±0,50) in the dry dock.
