Özet
Çalışmamızda Hedera helix folium ekstresi (Prospan®) tedavisinin sıçanlarda deneysel olarak oluşturulan adjuvan artrit modelindeki etkisini inceledik. Toplam 6 grup oluşturuldu. Birinci grup kontrol grubu olarak ayrıldı ve diğer grupların sağ arka ayak pençelerine 0. gün intradermal yolla 0,1 ml Freund’s Complete Adjuvant injeksiyonu yapılarak artrit oluşturuldu. 17-27 günler arası; ikinci gruba serum fizyolojik, üçüncü gruba Hedera helix folium ekstresi (Prospan®) (10 mg/kg), dördüncü gruba Hedera helix folium ekstresi (Prospan®) (30 mg/kg), beşinci gruba Hedera helix folium ekstresi (Prospan®) (50 mg/kg) ve altıncı gruba da pozitif kontrol için ibuprofen (100 mg/kg) uygulandı. Tüm ilaçlar sıçanlara gastrik gavaj yoluyla verildi. 0, 17, 20, 23 ve 27. günlerde pençe çapları kumpasla, ağrı düzeyleri ise basıç uygulama cihazıyla ölçüldü. 27. günde kalpten ponksiyonla kanlar alındı ve patolojik inceleme için sağ arka ayak eklemleri çıkarıldı. Serumda malondialdehit, interlökin- 1beta, interlökin-2, interlökin-6, interlökin-10 ve tümör nekroz faktör-alfa düzeyleri ölçüldü ve eklemler histopatolojik olarak incelendi. Hedera helix folium ekstresi (Prospan® 30 mg/kg) tedavisinin deneysel adjuvant artrit modelinde antiinflamatuvar etki göstererek pençe ölçüm değerlerini azalttığı, histopatolojik bulgulardan inflamasyon, vasküler proliferasyon, sinoviyal proliferasyon ve ödemde etkili olduğu görülmüştür. Ağrı parametre ölçümlerinde analzejik etkisi bulunamamıştır. Sitokin ölçümlerinde ise anlamlı sonuçlar elde edilememiştir.

In our study, we examined the effect of the Hedera helix folium extract (Prospan®) on the experimentally-induced adjuvant arthritis model in rats. In total, 6 groups have been formed. The first group served as the control group. In the other groups, 0.1 ml of Freund’s Complete Adjuvant was injected intradermally to the right back paws on day 0 to induce arthritis. Between days 17 and 27, saline has been administered to the 2nd group, Hedera helix extract (Prospan®) (10, 30 and 50 mg/kg) to the 3rd - 5th groups, respectively, and ibuprofen (100 mg/kg) was given as positive control to the 6th group. All of the drugs were administered through gastric gavage. On days 0, 17, 20, 23 and 27, paw diameters were measured with a caliper and pain was evaluated using a paw pressure apparatus. On day 27, bloods were taken from the heart by puncture and right hind leg joints were removed for pathological examination. Serum malondialdehyde, interleukin-1beta, interleukin-2, interleukin-6, interleukin-10 and tumor necrosis factor-alpha levels were measured and joints were examined histopathologically. Hedera helix folium extract (Prospan®) treatment exerted an antiinflammatory effect and reduced paw diameters in the experimental adjuvant arthritis model; histopathological findings indicate that it has been effective on inflammation, vascular proliferation, synovial proliferation and edema. No analgesic activity was observed in pain parameters. In cytokine measurements, no significant results were obtained.
