MMP-9 ve MMP-8 gen polimorfizmlerinin periodontitis ile ilişkisi

Özet
Periodontal hastalıklar genetik ve çevresel faktörlerin etkisiyle hiperemi, kanama, çep oluşum, diş kaybı ve klinik bağlantı epiteli kaybı gibi durumların gözlendiği multifaktöriyel bir hastalıktır. Patogenezinde özellikle genetik polimorfizmlerin ilişkileri rapor edilmiştir. Çalışmanın amacı, gen polimorfizmleri ile periodontal hastalık arasındaki korelasyonu değerlendirmek, genotip-fenotip ilişkisini ortaya koymak ve TNP'nin periodontitise yatkınlık u?zerindeki rolu?nu? incelemektir. MMP grubu ekstrasellu?ler matriks ile ilişkili olup periodontitis ve diğer hastalıklarda doku yıkım ve yenilenme metabolizmasının ana proteinazıdır. Çalışmamızda MMP-8 ve MMP-9 ile periodontitisin genetik ilişkisinin incelenmesi için hastalardan alınan tu?ku?ru?k örneklerinden genomik DNA'lar izole edilerek PCR ve RFLP yöntemleriyle MMP-9 (rs17576) Q279R ve MMP-8 (rs1940475) Lys87Glu gen polimorfizmi araştırılması planlanmıştır. Trakya Üniversitesi Diş Hekimliği Faku?ltesi Periodontoloji Ana Bilim Dalı'ndan 100 periodontitis hastası ve 100 periodontal durumu sağlıklı gönu?llu? toplamda 200 akrabalık ilişkisi olmayan Tu?rk hasta çalışmaya dahil edilmiştir. Çalışmamızın sonuçlarına göre MMP-8 ve MMP-9 ile ilişkili bakılan gen bölgelerinin periodontitis için genotip ve allel karşılaştırmasında istatistiksel olarak belirgin farklılık bulunmamıştır (p <0,05). Periodontal diseases are multifactorial diseases in which genetic and environmental factors affect the clinical outcomes of the disease such as hyperemia, bleeding on probing, pathological pocket formation, tooth loss, and clinical attachment loss. Especially genetic polymorphisms have been previously reported in the pathogenesis of chronic periodontitis. The study aims to evaluate the correlation between gene polymorphisms and periodontal disease, to reveal the genotype-phenotype relationship, and to examine the role of SNP on predisposition to periodontitis. MMPs are associated with extracellular matrix metabolism and the main proteinases related to tissue destruction and remodeling events in periodontal and other diseases. Consequently, to examine the genetic relationship between MMP-8, MMP -9, and periodontitis, genomic DNAs were isolated from saliva samples that were obtained from patients. The Q279R (rs17576) and Lys87Glu (rs1940475) gene polymorphism were studied by PCR and RFLP methods. A total of 200 unrelated Turkish subjects including 100 Periodontitis patients and 100 subjects with healthy periodontal conditions were recruited from the Department of Periodontology, School of Dentistry, Trakya University. According to the results of our study, statistically, there were no significant differences could be found between genotypes or allele frequencies of the gene regions associated with MMP-8 and MMP-9 for periodontitis (p <0,05).
