Deneysel olarak oluşturulmuş meme tümöründe tamoksifenin lipid ve lipoproteinler üzerine etkisinin incelenmesi

Özet
Bu araştırmada, deneysel olarak farelerde solid meme kanseri oluşturularak tamoksifen tedavisinin lipid, lipoproteinler ve lipid peroksidasyonu üzerine etkileri incelendi. Denekler, 4 gruba ayrıldı. Tedavi 21 gün sürdü. Tamoksifen, zeytinyağı içinde çözündürülerek gavaj ile verildi. Serumda total kolesterol, trigliserid, yüksek dansiteli lipoprotein kolesterolü, çok düşük dansiteli lipoprotein kolesterolü ve malondialdehit düzeyleri ölçüldü. Total kolesterol düzeyinin, hem sağlıklı hem de tümörlü kontrol grubuna göre; tamoksifen tedavisi verilen gruplarda anlamlı olarak azaldığı gözlendi. Trigliserid düzeyinin, sağlıklı kontrol grubuna göre; tümörlü grupta arttığı, tamoksifen tedavisinin anlamlı düşüşe yol açtığı bulundu. Yüksek dansiteli lipoprotein kolesterolü düzeyinin tümörlü gruplarda anlamlı olarak azaldığı, bu azalmanın tamoksifen tedavisi ile daha da arttığı bulundu. Çok düşük dansiteli lipoprotein kolesterolü düzeyinin, sağlıklı kontrol grubuna göre; tümörlü kontrol grubunda arttığı, tamoksifen tedavisi ile anlamlı olarak azaldığı gözlendi. Malondialdehit düzeyinin ise sağlıklı kontrol grubuna göre; tümörlü kontrol grubunda, düşük doz tamoksifen tedavisi verilen grupta, yüksek doz tamoksifen tedavisi verilen grupta anlamlı olarak artmış olduğu gözlendi. Malondialdehit düzeyinde görülen yükselme en çok yüksek doz tedavi grubunda idi.
Abstract
In this research, solid breast cancer is formed experimentally in mice and the effects of tamoxifen treatment on lipids, lipoproteins and lipid peroxidation are examined. The mice were grouped into 4. The duration of treatment was 21 days. Tamoxifen was dissolved in olive oil and given by gavage. The levels of total cholesterol, triglyceride, high density lipoprotein cholesterol, very low density lipoprotein cholesterol and malondialdehyde in serum were measured. The levels of total cholesterol were found to be significantly decreased in tamoxifen treatment groups when compared with both healthy control group and control group with tumors. Triglyceride level was found to be increased in group with tumors when compared with healthy control group. Tamoxifen treatment caused statistically significant decrease. The levels of high density lipoprotein cholesterol were found to be significantly decreased in groups with tumors. This decrease increased more with tamoxifen treatment. The level of serum very low density lipoprotein cholesterol was found to be increased in group with tumors, when compared with the healthy control group. The level decreased significantly with tamoxifen treatment. Serum malondialdehyde levels were measured to be increased significantly in control group with tumors, low dose tamoxifen treatment group and high dose tamoxifen treatment group when compared with healthy control group. The maximum increase in malondialdehyde level was in high dose group.
