Öğe Ameliorative effects of melatonin on intestinal oxidative damage in streptozotocin-induced diabetic rats(Istanbul Univ, Fac Pharmacy, 2020) Yakut, Zatiye Ayca Cevikelli; Akcay, Gizem Buse; Cevik, Ozge; Sener, GokselBackground and Aims: The potential therapeutic effects of melatonin on changes in intestinal tissue of diabetic rats were investigated. Methods: Male Sprague-Dawley rats were assigned into 5 groups (10 rats in each): Control, diabetes, diabetes+insulin, diabetes+melatonin, and diabetes+insulin+melatonin groups. Streptozotocin (60 mg/kg) was administered intraperitoneally to the rats to induce diabetes. At the end of 8 weeks of treatment, after blood glucose measurement and subsequent decapitation, glutathione (GSH) and matondialdehyde (MDA) levels and caspase-3, myeloperoxidase (MPO), and superoxide dismutase (SOD) activities in the intestinal tissue were investigated. Results: in diabetic animals, elevated blood glucose levels caused oxidant damage in the intestinal tissue that was demonstrated with increased MDA levels, caspase and MPO activities, and decreased GSH levels and SOD activities. Although melatonin demonstrated more significant results than insulin, separate administration of both melatonin and insulin improved the oxidative damage parameters compared to the diabetes group. In the combined treatment group, all parameters were back to control levels statistically more significant when compared with the treatment-alone. Conclusion: Melatonin has been shown to protect intestinal tissue from diabetic oxidant damage. With insulin treatment in type I diabetes, melatonin supplements may increase the quality of life through reducing complications.Öğe BIOLOGICAL ACTIVITY EVALUATION OF THREE Hypericum L. SPECIES(Trakya Univ Balkan Yerlesesi Enstituler Binasi, 2022) Eruygur, Nuraniye; Ozpinar, Necati; Ozpinar, Hulya; Atas, Mehmet; Tekin, Mehmet; Cevik, OzgeThis study was performed in order to evaluate in vitro antioxidant, anti-Trichomonas vaginalis, antimicrobial and cytotoxic activities of methanol extracts of aerial parts of Hypericum perforatum L., H. thymbrifolium Boiss. & Noe and H. thymopsis Boiss. The antioxidant activity of methanol extracts was tested with 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2 '-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging and iron chelating assays. The total phenol and flavonoid contents were also determined spectrophotometrically. Cytotoxicity and apoptosis were evaluated on MCF-7 breast cancer and PC3 prostate cancer cell line. Antimicrobial activities were evaluated by the microdilution method on the bacterial strains Staphylococcus aureus (ATCC 29213), Enterococcus faecalis (ATCC 29212), Pseudomonas aeruginosa (ATCC 27853) Escherichia coli (ATCC 25922) and the fungal strain Candida albicans (ATCC 10231). All extracts demonstrated cytotoxic activity on PC3 prostat cancer cell line with a concentration dependent manner, while H. thymopsis extract was the most active against cancer cell growth. The obtained results of the present study revealed that the methanol extract of H. perforatum, H. thymbrifolium and H. thymopsis have significant antioxidant and cytotoxic activities. The results provided the basic research data for further phytochemical and biological activity guided investigations on these species for identication and isolation of potential drug active compounds.Öğe The effects of riboflavin on ischemia/reperfusion induced renal injury: Role on caspase-3 expression(Marmara Univ, 2019) Ayaz Adakul, Betul; Ertas, Busra; Cevikelli, Zatiye Ayca; Ozbeyli, Dilek; Ercan, Feriha; Kandemir, Cansu; Cevik, OzgeReactive oxygen metabolites play important roles in ischemia/reperfusion (I/R) injury in several organ systems. Riboflavin has been shown to exert antioxidant and/or anti-inflammatory activities in several experimental models. The aim of this study was to investigate the role of riboflavin against I/R injury in the rat kidney. Wistar albino rats 200-300 g weighing were divided into 3 groups. One week after unilateral nephrectomy, the IR procedure was applied to the rats. To induce I/R injury renal pedicle was clamped for 45 minutes and then rats were allowed reperfusion for 6 hours. Riboflavin (25 mg/ kg, orally) or vehicle was administered for one week as pretreatment. After decapitation, kidney tissue samples were taken for the evaluation of malondialdehyde (MDA), an end product of lipid peroxidation; glutathione (GSH), a key antioxidant; and 8-hydroxydeoxyguanosine (8-OHdG), a specific marker of oxidative DNA damage. Furthermore, myeloperoxidase (MPO) and caspase-3 activities were also examined together with histological analysis. Ischemia/reperfusion induced significant increases in MDA and 8-OHdG levels and MPO and caspase- 3 activities, and decrese in GSH levels. In the riboflavin treatment these indices were found to be reversed back to control levels. The present data demonstrated that riboflavin, through its antioxidant effect, attenuates I/R induced acute renal injury in rats.Öğe Evaluation of in vitro Antioxidant, Antimicrobial and Cytotoxic Activities of Crude Ethanol Extract and Fractions of Achillea sintenisii Hub. Mor.(Marmara Univ, Inst Health Sciences, 2023) Eruygur, Nuraniye; Atas, Mehmet; Tekin, Mehmet; Cevik, OzgeObjective: The Achillea species have been used to treat various ailments due to its anti-inflammatory, hemostatic, spasmolytic and cholagogue effects in the Turkish traditional medicine. However, there is no biological activity studies on some Achillea species except for the well-knowns. This work aimed to determine the antioxidant, antimicrobial and cytotoxic activities of the crude ethanolic extracts and fractions of Achillea sintenisii using in vitro methods. Methods: The antioxidant activity was investigated by DPPH (1,1'-diphenyl - 2-picrylhydrazyl), ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging, total phenol and total flavonoid content, and iron chelating methods. Antimicrobial activity evaluated by micro-plate dilution method against five test organisms. Cytotoxicity was determined by MTT method using MCF-7 breast cancer cell line and PC-3 prostate cancer cell line. Apoptosis was also measured by AO/EB staining. Results: The n-Hexane fractions showed the highest antimicrobial and cytotoxic activities, respectively. Administration of the extracts on the cancer cells showed a concentration dependent inhibition on cell proliferation. The anti-proliferation effect could be via apoptosis and associated with the cell death. Conclusion: The results showed that the extracts demonstrated antioxidant, antimicrobial, and cytotoxic activity, also supports the claims of traditional usage.Öğe Exercise and caloric restriction improve cardiovascular and erectile function in rats with metabolic syndrome(Springernature, 2021) Cevikelli-Yakut, Zatiye Ayca; Ozcelik, Reyhan; Cevik, Ozge; Sener, Tarik Emre; Sener, GokselThe aim of this study is to examine the possible benefits of exercise and caloric restriction (CR) on cardiovascular hemodynamics, erectile function, and antioxidant system in metabolic syndrome (MS). Sixty male Spraque-Dawley rats were divided into five groups; control, MS, MS + CR, MS + exercise (EXC), and MS + CR + EXC. To induce MS, 10% fructose solution was applied for 3 months. Thereafter, in CR groups calorie was restricted 40% and in EXC groups swimming was performed for 6 weeks. Body weight, blood glucose, and blood pressure (BP) levels were measured before and after MS induction and at the end of the experiment. After decapitation, tumor necrosis factor (TNF)-alpha, adiponectin (ADP), and plasminogen activator inhibitor (PAI)-1 levels were investigated in blood, oxidative stress parameters were examined in heart, aorta, and corpus cavernosum (CC) tissues. Isometric contraction in isolated tissue bath was studied in aorta and CC tissues. Animals subjected to exercise and CR had decreased BP and blood glucose levels. Impaired contraction-relaxation responses in MS group were improved with exercise and CR. MS-induced increase in TNF-alpha, PAI-1, malondialdehyde (MDA), and decrease in ADP, glutathione (GSH), and superoxide dismutase (SOD) were normalized with exercise and CR. Exercise and CR may be beneficial against changes in cardiovascular hemodynamics caused by MS.
